SUMMARY We studied the respiratory burst activity of peripheral blood monocytes and polymorphonu clear leukocytes (PMN) in eleven patients with psoriasis and eleven healthy controls, using zymosan particles to activate the phagocytes in vitro. The activation of the phagocytes was measured in a luminol-enhanced chemiluminescence (CL) assay. We found a significantly higher respiratory burst activity of both monocytes and PMN in patients with psoriasis compared with the control subjects. The percentages of monocytes in psoriatics did not differ from those of the controls. Evaluation of remission in adult acute lymphoblastic leukemia (ALL) normally relies on cytologic evaluation and flow-cytometric analysis. The diagnostic impact of bone marrow trephine biopsies has not been studied so far. We investigated 26 biopsies of 16 patients with T-ALL. Double fluorescent immunostaining with TdT and CD3 was performed. Corresponding cytologic evaluation and flow-cytometric data were available. In 17 of 26 investigations, the results were concordant (either positive or negative). In one examination, residual blasts were not recognized by trephine biopsy, but by other methods. High resolution analysis and differential condensation in RBA-banded human chromosomes.

Human prophase, premetaphase, and mid-metaphase chromosomes are prepared and analyzed using the thymidine cell synchronization technique and R-banding patterns (RBA). Haploid sets with 700–1000 bands can be demonstrated. Sequences of chromosomes of different degrees of condensation are helpful for a better understanding and classification of regions of extended chromosomes. A considerable variation in the condensation of parts of homologous chromosomes is reflected in the variability of the arm ratio. This differential condensation of chromosomes is entirely effected by variation of the degree of condensation in AT rich interbands and can be attributed to the degree of labeling by BrdU.

The single cell gel test (SCG-test or comet assay) is a rapid and sensitive method for measuring DNA damage and repair in individual cells. A wide variety of mutagens have been shown to cause DNA alterations detectable with the comet assay, but it is not yet clear whether a relationship exists between the DNA effects and the induction of mutations.
